Cloning and Identification of A Novel Murine Organic Anion Transporter like Protein 2 in Kidney

Abstract
The organic anion transporters (OATs)are expressed in various tissues, primarily kidney and liver, but also in placenta, small intestine, and choroid plexus, which are all epithelial tissues that transport xenobiotics. Until now six isoforms of OATs and a organic anion transporter like protein (OATLP) have reported. Considering variety of organic anionic compounds, the existence of other OATs¡¯ isoform was assumed. In this connection, we have searched new isoform in expressed sequence tag (EST) database. We found a candidate of new clone (BC021449) from mouse kidney cDNA library and named mouse OAT like protein 2 (mOATLP2). The mOATLP2 cDNA consisted of 2016 base pairs that encoded a 551 amino acid residue protein with 12 putative transmembrane domains. The deduced amino acid sequence of mOATLP2 showed 37 to 63% identity to those of other members of the OAT family. The tissue distribution using Northern blot analysis, mOATLP2 expressed predominantly in kidney. When the investigated Western blot analysis using synthesized oligopeptide-induced mOATLP2 antibody, about 90-kDa band was detected. From these results, mOATLP2 may become a novel candidate of organic anion transporter isoform identified in the kidney.
